PRIMERO is a longitudinal birth cohort which aims to identify the underlying factors associated with the development of asthma in healthy minority children. With the height of the pandemic, PRIMERO also provides a prime opportunity to study the effects of SARS-CoV-2 and related stresses on the mother and infants with admixed ancestry. The PRIMERO study is a cooperative agreement with NIHLBI (U01HL) involving a funding of nearly $10 million to recruit 3000 healthy infants within a three-year window and follow these healthy infants to age 5. Funded in 2020, PRIMERO is actively recruiting to meet the goals of the U01. In 2020 (year 1 of U01), PRIMERO has enrolled over 400 subjects from March to December, with 2600 recruits pending. Every mother-child dyad pair will have biospecimens collected. Besides DNA, RNA, and plasma extracted from the mother’s peripheral blood, cord blood will be collected from every infant recruited into the study. Plasma, DNA and RNA will be extracted from the cord blood and CBMC (cord blood mononuclear cells) will be cryopreserved. All biospecimens will be banked, logged into a database and stored for future analysis. Given the rapid denaturation of the biospecimens, the samples must be processed in a timely fashion to maintain sample integrity and viability. The successful applicant will be assisting with storing and recording the extraction of DNA, plasma, and serum from cord and peripheral blood for further analysis. In ition, the LA I will be assisting staff with and learning about basic molecular biology related experiments and other miscellaneous everyday lab functions.
