A proper understanding of these drug substances and drug products relies on innovative analytical techniques including NMR structure elucidation. We wish to achieve a better understanding of factors that define drug substance properties, and use this understanding to positively influence the steps needed to produce the final drug product formulation. Several of the vaccine drug products in development consist of polysaccharides connected to proteins. This development would benefit from a better understanding of the chemical processes used in the conjugation of polysaccharide chains to carrier proteins. Knowledge built in this space would potentially impact experimental approaches to optimizing the reaction conditions for vaccine production. Another example is the characterization of antibodies using isotopic labeling and multidimensional NMR methods to understand reactivity and dynamics of antibody based pharmaceutical products. The proposed research will cover the development of methods for the analytical characterization of a) polysaccharide serotypes and their covalent coupling to carrier proteins (e.g. CRM197) and b) antibodies and their covalent modifications focusing on Fab and Fc subdomains. n nWe are looking for an independent and scientifically driven candidate that will be responsible for the design and execution of protein-based NMR experiments, such as 2 and nD heteronuclear correlation and 2D dynamics pulse sequences. These methods will be used to determine the assignment, reactivity and relaxation properties of pharmaceutically relevant molecules. You should have a passion for biophysics and would ideally be able to analytically characterize biomolecules using biophysical methods beyond NMR such as, but not exclusively, calorimetry or mass spectrometry. Expertise in working with 3D structures will benefit the role as you work together with chemists, biochemists and structural biologists to build hypotheses around structures of biomolecules and their complexes. We are looking for a highly skilled and motivated individual, who, while capable of working independently, understands and values cross-functional teamwork and will support our common mission in the invention and development of analytical methods for new therapeutic solutions to unmet medical needs. You will collaborate with our team and chemists across the organization. n nWhat You Need: n nCandidates must currently hold a PhD OR receive a PhD by fall of 2021. n nPreparative and analytical biochemical and biophysical methods (chromatography, electrophoresis, Surface Plasmon Resonance, Calorimetry). n nExperience in the NMR of biological macromolecules. n nExperience in the application of computational methods for the assignment of proteins based on multidimensional NMR methods. nnIt’s Great If You Have: n nExperience in synthetic chemistry. n nComputational skills: knowledge of DFT, MD and distance geometry software. n nExperience in molecular structure calculations. n nAn understanding to protein structure, function and dynamics. nWe are a research-driven biopharmaceutical company.
