Primary Responsibilities: We are looking to fill an opening for an entry level Research Associate to support the Discovery Biology group, reporting to a Senior Scientist. The position will support various personnel within the group and involve diverse technical responsibilities. While the expectation is that training will be necessary for many aspects of the position, some prior experience in a lab, either full time or undergraduate part time work, is essential. The role will support numerous efforts within Discovery Biology, encompassing antibody discovery, humanization and optimization, ligand and receptor biology, as well as assays for lead identification and selection. In ition, the role may also involve research into molecular mechanisms underlying lead candidates to support advancement of drug candidates. This is a research role, and will require reading and understanding of scientific publications on cutting edge biology, and an independent drive to understand a changing landscape. Follow standard lab procedures to generate molecular biology reagents for group use. Clone, modify, and generate plasmid vectors for protein production, cell line generation, and other cell based assay use. Routinely enter materials into a laboratory inventory management and tracking system, and update as necessary. Including plasmids, preps, cell lines, and purified proteins. Perform single cell isolation of antibody secreting cells for antibody discovery efforts, and assist in antibody gene amplification and downstream screening. Carry, transfect, and provide mammalian culture lines for research scale protein production and cell based assays, and analyze samples as required. Maintain an up to date, meticulous laboratory notebook describing all lab work. Order lab supplies, and generate consumables such as plates and competent cells as needed for the group. Skills Experience with: Molecular cloning, and commonly used techniques such as: PCR, fragment assembly, site directed mutagenesis, restriction digestion, transformation, colony screening, gel electrophoresis, and primer design. The use of vector design software and sequence alignment. Kit-based column nucleic acid purification: maxipreps, minipreps, etc. Familiarity with: Fundamental protein assays such as: Western Blots, ELISAs, protein quantification (Bradford/BCA). Mammalian tissue culture, including passaging, transfection, and harvesting. Microsoft Word/Excel/Powerpoint, GraphPad Prism, Snapgene. Pubmed, NCBI, uniprot, ensembl. Experience B.S. degree in molecular biology/biological sciences or a related discipline with 0-3 years postgraduate experience.
