Research Technologist GENERAL SUMMARY/ OVERVIEW STATEMENT: The Research Technologist in the Molecular Neurogenetics Unit in Bldg 149 is a full-time position under the direct supervision of the PI or Lab Director. This technologist independently carries out a broad range of technically advanced research activities and procedures; evaluate conclusions and has considerable latitude to modify or devise methods and techniques as necessary to achieve desired results; and trains and supervise technical staff and students in the research unit. Must be able to oversee and supervise employees as well as function as a team member. Must be able to act as team leader in projects, write up results for publication, and prepare images for talks given by the PI. Experience in handling mice, cell culture and molecular biologic methods is required. PRINCIPAL DUTIES AND RESPONSIBILITIES: Performs duties of Research Technologist plus: – Directs and carries out experiments independently on projects with collaborators with the advice of the PIs – Trains and provides ongoing guidance for technicians and students in use of laboratory equipment, including Nanosight, Bioanalyzer, PCR machine, and ultracentrifuge – Generates and maintains a book of laboratory protocols for Unit use. – Independently performs cell and molecular biology experiments, including: cell culture; cell transfection with DNA constructs and cell transduction with AAV and lentvirus vectors; purification and analysis of minute amounts of RNA/DNA; evaluation of RNA quality using the Bioanalyzer; ELISA; immunocytochemistry; fluorescence microscopy; western blots and antibody biotinylation – Independently performs and trouble shoots experiments on isolation and analysis of extracellular vesicles (exosomes), including: vesicle isolation using differential centrifugation and density gradients, as well as magnetic beads using streptavidin and biotinylated antibodies; quantification and sizing of vesicles using the Nanosight; and developing novel methods for analysis, including bioluminescence measurements – Carries out mouse experiments including: mouse breeding, genotyping and timed pregnancies; drug administration; surgeries, injections tissue and blood harvesting and cryosectioning; preparation of primary cell cultures from mouse embryos of neurons, microglia, fibroblasts and mesothelial cells; and generation of single cell suspensions from adult mouse brain for FACS analysis. – Participates in the design and modification of research protocols and experimental design – Develops research methodologies within the parameters of experimental protocols and research objectives. – Responsible for oversight and coordination of scheduling of experimental procedures. – Orients and train new research technicians as needed – Responsible for troubleshooting problems and instructing others in highly specialized techniques. – May develop modifications of existing assay techniques, experimental protocols, or new procedures. – Synthesizes and interprets results. – Performs advanced data analysis using advanced statistical techniques as needed – Prepares written and/or verbal status reports as needed, including help in writing of papers and preparation of images for talks – Uses statistical and graphic summaries. – Composes and may present sections of research reports and manuscripts as needed – Maintain records in accordance with strict laboratory practice guidelines – Assist with other core work as needed, such as: handling and distribution of reagents, lab organization, monitoring of all equipment – Expertise in computer programs used by the Unit. SKILLS & COMPETENCIES REQUIRED: – Ability to make independent effective decisions and trouble shoot instruments – Demonstrated competence in advanced research techniques and methodologies – Strong organizational and communication skills. – Ability to supervise and train staff effectively. – Strong database management and over all computer skills. – Demonstrated analytical skills to problem solve effectively. – Functions with speed, precision, and an understanding of theory behind tests and procedures EDUCATION: Bachelor’s Degree EXPERIENCE: Required: Minimum 3 years of directly related and progressively more responsible experience required. SUPERVISORY RESPONSIBILITY (authority to hire, promote, or terminate): Will train and supervise Lab Assistants, Technicians, Technologists and Graduate Students. FISCAL RESPONSIBILITY: May prepare and monitor budget for research supplies. WORKING CONDITIONS: Duties will be performed in a research laboratory. Work will be conducted over a forty-hour workweek.
